PD98059, SB203580, and Akt-deficient mice reduced stretch-induced Akt and mitogen-activated protein kinase activation. After 5 days of bleomycin administration, wild-type mice or serine/threonine kinase-protein kinase B (Akt)+/- mice ventilated at a tidal volume of 6 ml/kg or 30 ml/kg for 1 hour were pretreated with 2 mg/kg PD98059 or 16 mg/kg SB203580 subcutaneously for 30 minutes. Western blot analysis was performed using an antibody that recognizes phosphorylated Akt expression, extracellular signal-regulated kinase (ERK) 1/2 expression, or P38 expression ((a) to (c) top panel), and an antibody that recognizes total Akt, ERK1/2, or P38 protein expressions in lung tissue ((a) to (c) middle panel).
