Optimal reaction temperature for the PA-MCDA primer assay. MCDA reactions when detecting the P. aeruginosa gene were monitored by real-time measurement of turbidity. A turbidity of > 0.1 was considered positive. Nine kinetic graphs were obtained at various temperatures (59–67 °C, at 1 °C intervals) with P. aeruginosa DNA at a concentration of 1 ng per tube. The graphs showed that 65 °C was an optimal temperature for amplification
