(a) Monocytes were gated using a forward scatter/side scatter plot and a CD14/CD33 scatter plot, where CD33^negative cells (lymphocytes and plasmacytoid dendritic cells (PDCs)) as well as CD14^negativeCD33^bright myeloid dendritic cells (MDCs) were excluded from further analysis. According to their differential expression of CD14 and CD16, three monocyte subsets were defined: CD14^brightCD16^negative, CD14^brightCD16^positive and CD14^dimCD16^positive. Proportions and cell counts of each subset are given. (b) Dendritic cell subsets were defined as follows: PDCs were gated as lineage marker (CD3, CD14, CD16, CD19, CD20, CD56)-negative (lin^-) CD123⁺human leukocyte antigen (HLA)-DR⁺ cells, and MDCs as lin ^-CD33⁺HLA-DR⁺ cells.
