Anti-PD-1 increases IFN-γ+ CD4 and CD8 T cells in fungal sepsis. Mice had tail-vein injection of Candida and were treated with anti-CTLA-4 or anti-PD-1 as described in Figure 4. Following supernatant harvesting, splenocytes were treated with brefeldin for four hours and then fixed, permeabilized and immunostained for CD4, CD8 and intracellular IFN-γ.Flow cytometry was performed to quantitate the percentage of CD4 and CD8 T cells that were positive for IFN-γ.Anti-PD-1 but not anti-CTLA-4 increased the percentage of IFN-γ+ CD4 and CD8 T cells, (P <0.05).
